Motorists are being warned to avoid the main road from Ballacraine to Kirk Michael after police closed it following an accident.
The serious incident, which happened shortly after 7am, involves a motorbike and officers say the road may be closed for some time.
Drivers are being asked to avoid the area until further notice.
Bus Vannin, meanwhile, says buses - including school services - will divert via the coast road to Peel and St John's until at least 11:30am.
